چکیده
background: creation of artificial gametes may provide a universal solution for these patients of lacking gametes. stem cell technology may provide a way to obtain fully functional gametes. retinoic acid (ra) can initiate meiosis. several studies have demonstrated that ra can promote sperm cells differentiation from mouse embryonic stem cells (mescs) and other cells from human embryonic stem cells (hescs). objective: we sought to determine whether ra could promote differentiation of germ cells from hescs. materials and methods: hescs were differentiated as embryoid bodies (ebs) in suspension with all-trans ra (atra) or without atra for 0, 1, 3, 5 and 7 days, and then compared the expression of vasa, scp3, gdf9 and tekt1 by real-time pcr. the statistical differences were evaluated by one way anova. results: the expression of germ cell-specific markers including the gonocyte marker vasa, the meiotic marker scp3, and postmeiotic markers, gdf9 and tekt1, all increased in the presence and absence of ra as eb differentiation progressed. in addition, the expression of these markers increased an average of 9.3, 6.9, 7.2 and 11.8 fold respectively in the presence of ra, compared to the absence of ra, over 5 days differentiation. conclusion: our results indicate that hescs may have the potential to differentiate to primordial germ cells (pgcs) and early gametes. and ra can improve germ cells differentiation from hescs.

Retinoic acid improve germ cell differentiation from human embryonic stem cells
BACKGROUND Creation of artificial gametes may provide a universal solution for these patients with no gametes. Stem cell technology may provide a way to obtain fully functional gametes. Retinoic acid (RA) can initiate meiosis. Several studies have demonstrated that RA can promote sperm cells differentiation from mouse embryonic stem cells (mESCs) and other cells from human embryonic stem cells ...
متن کاملAnalysis of Promyelocytic Leukemia in Human Embryonic Carcinoma Stem Cells During Retinoic Acid-Induced Neural Differentiation
Background: Promyelocytic leukemia protein (PML) is a tumor suppressor protein that is involved in myeloid cell differentiation in response to retinoic acid (RA). In addition, RA acts as a natural morphogen in neural development. Objectives: This study aimed to examine PML gene expression in different stages of in vitro neural differentiation of NT2 cells, and to investigate the possible role o...
متن کاملThe effect of BMP4 on mouse embryonic stem cell proliferation and differentiation into primordial germ cells
Background and Aim: Artificial gamete production from stem cells is a novel strategy for treatment of infertility. Among various stem cell sources, embryonic stem cells (ESC) can be considered as an appropriate source for in vitro formation of germ cells. In this study we evaluated the effect of BMP4 on proliferation and differentiation of mouse embryonic stem cells into primordial germ cells (...
متن کاملCHIR99021 combined with retinoic acid promotes the differentiation of primordial germ cells from human embryonic stem cells
Primordial germ cells (PGCs) derived from human embryonic stem cells (hESCs) represent as a desirable experimental model as well as a potential strategy for treating male infertility. Here, we developed a simple and feasible method for differentiation of PGCs from hESCs by using CHIR99021 (an inhibitor of glycogen synthase kinase 3) and retinoic acid (RA). We firstly found that the deleted in a...
متن کاملDazl promotes germ cell differentiation from embryonic stem cells.
It has been demonstrated that through the formation of embryoid bodies (EBs) germ cells can be derived from embryonic stem (ES) cells. Here, we describe a transgene expression approach to derive germ cells directly from ES cells in vitro without EB formation. Through the ectopic expression of Deleted in Azoospermia-Like (Dazl), a germ cell-specific RNA-binding protein, both motile tailed-sperm ...
متن کاملProliferation in culture of primordial germ cells derived from embryonic stem cell: induction by retinoic acid
An in vitro system that supports primordial germ cells (PGCs) survival and proliferation is useful for enhancement of these cells and efficient transplantation in infertility disorders. One approach is cultivation of PGCs under proper conditions that allow self-renewal and proliferation of PGCs. For this purpose, we compared the effects of different concentrations of retinoic acid (RA), and the...
